    PolyLC do PRIONS

    Recent outbreaks of bovine spongiform encephalopathy (BSE) or mad cow disease in Europe have highlighted the need to develop a method to identify pathogenic prion proteins in the blood of animals and humans.

    In collaboration with Dr. Mary Jo Schmerr of the United States Department of Agriculture (USDA), PolyLC has helped to develop the first practical test for pathogenic prion proteins in blood.  Experimentally, the test has given positive results in a variety of TSE (Transmissible Spongiform Encephalopathy) diseases, including:
     

    • Scrapie (in sheep)
    • Chronic wasting disease (in elk and mule deer)
      and similar prion diseases in hamster, llama, chimpanzee and humans


    At present the test can detect prion protein in the picogram range per ml. blood.  This is sufficiently sensitive to give positive results in infected animals before clinical symptoms develop.  The test has not yet been validated. Validation studies are in progress in the case of scrapie.  That will require correlation of the positive results with the animals subsequently developing clinical cases of disease.  This should take 1-3 more years.  In the interim, all use of this test should be considered to be research rather than definitive diagnosis.

    PolyLC’s HPLC columns of PolyHYDROXYETHYL Aspartamide  permit the isolation of pure pathogenic prion protein from extracts from brain and other tissues of diseased animals.  The protein is obtained free of detergent with recovery >90%, in a form that readily forms complexes with anti-prion antibodies
